Abstract
  • The bacterium most frequently used in microbial leaching processes and studies is Thiobacillus ferrooxidans. Applications of this bacterium include the treatment of coal, low-grade ores and industrial solid wastes for the solubilization of pyrite and other trace metals associated with sulfides. The goal of this study was the development of a standard procedure that can be used for screening or comparing various strains of Thiobacillus ferrooxidans. Measuring the oxidation of ferrous iron is a quick and simple method for predicting the relative effectiveness of a given strain of T. ferrooxidans in microbial leaching operations. When conducting experiments for the relative determination of bacterial activity, 9K medium with a low initial soluble ferrous iron concentration and a pH of approximately 2.0 is recommended. Also it is important to begin with the same ferric iron concentration in bacterial cultures, while minimizing the amount of precipitated iron. The effect of inoculum size on the maximum oxidation rate of ferrous iron is negligible; however, lag time can be reduced by using larger inocula.
