Abstract
  • The Micralyne microfluidic toolkit (μTK) was an instrument built by Micralyne in the early 2000’s. It was built for electrophoresis and has a sensitive laser-induced fluorescence detection system. The updates to the system were upgrading the control GUI from running in LabVIEW 5 (Windows XP, 2000) to LabVIEW 2012 (Windows Vista, 7) and repairing the high voltage board. Along with that, the electrophoretic and detection capabilities of the μTK were confirmed and settings for the voltage and focusing established. Phosphate buffer solution was used as both a pH buffer and as a source of ions to establish an electric field for electrophoresis; all solutions were phosphate buffer solutions at pH 7. Rhodamine B was the fluorophore of choice used as the analyte for fluorescence detection and as the background for inverse fluorescence. Caffeine and acetaminophen were the analytes for inverse fluorescence. Fluorescence detection was successfully demonstrated with Rhodamine B as an analyte. While inverse fluorescence detection was not demonstrated with success, a path forward to achievement of this goal was demonstrated. Key Words: Electrophoresis, Fluorescence, LabVIEW
