Abstract
  • The purpose of this project is to characterize mixing effects of the Beta 2 tool that is under development by the Hewlett Packard Pinot team. By optimization of a stage oscillation process, the undesirable effects of a locally high concentrated drug elution within a well plate can be alleviated. This requires quantitative measurement of the mixing associated with “jitter” from the Beta 2 tool for both 96 and 384 well plates. This project began by researching theoretical underpinnings of mixing within the well plates and calculating dimensionless parameters associated with mixing from jitter and well size. After this, the team conducted a number of experiments with high and low factors to determine the optimal settings for mixing. These factors included stage motion (axis and time), volume dispensed, jitter mode, and amplitude (jitter distance). It has been determined that jittering post dispense is the optimal mixing method for both large and small dispense volumes in the 96 well plates. It is also recommended to move the stage in both the x and y direction for a minimum of 3 seconds at an amplitude of 1.25 mm. It has also been concluded that the instrument cannot mix the wells of 384 well plates.
