The Relationship Between Gestational Diabetes and the Risk of Orofacial Clefts in Infants: A Systematic Review

  • Background: This study investigated the relationship between gestational diabetes mellitus (GDM) and orofacial cleft (OFC). The association between GDM and OFC has been debated, therefore this systematic review focused on presenting a synthesis of the previous studies in this field. Methods: PubMed was searched using MeSH terms for articles published through February 28th, 2021. Retained studies were then examined to retrieve quantitative and qualitative data to draw comparisons and analyze the association between GDM and OFC. Results: Among the 418 articles that were obtained from PubMed, eight articles met the inclusion criteria. The retained articles included four case-control studies, three cohort studies, and one cross-sectional study. Four of the studies reported the presence of a weak association between GDM and OFC (OR range: 1.40-1.60), while the other four studies reported no association between the two variables. Three of the four case-control studies reported weak, but significant associations. In contrast, only one cohort study reported an association between GDM and OFC, but only when mothers with GDM were also obese. Conclusion: This review found no evidence of an association between GDM and OFC. This study highlights the lack of research conducted on the relationship between these variables and calls for further investigative studies.
