Abstract
  • The detection of mammalian FMO’s is important in the understanding of active FMO 2.1 and its role in metabolizing drugs to treat tuberculosis. Refinement of an absolute quantification (AQUA) mass spectrometry method is needed to be able to quantify the different FMO isoforms present in mammalian tissue. We are doing studies with wildtype and Fmo 1/2/4 null mice as a model for human FMO2.1 and FMO2.2, respectively and used model mouse Fmo peptides for our studies. Following denaturing polyacrylamide gel electrophoresis, an in-gel trypsin digestion was performed with labeled and unlabeled FMO proteins present. The samples were run on LC/MS/MS. Both the labeled and unlabeled FMO proteins from in-gel digestion were detected by LC/MS/MS along with a serial dilution of, directly injected, labeled FMO peptides.
