  • Auxotrophic strains of the fungus Fusarium graminearum were generated in order to increase the number of functional selectable markers and improve the utility of the model organism. Single metabolic genes, the sequences encoding for enzymes required for the production of specific essential metabolites, were deleted in F. graminearum, and then validated strains were crossed to generate single strains with multiple metabolic gene deletions. To demonstrate the function and efficacy of these auxotrophic strains for future genetic analysis, single gene auxotrophs were complemented with the respective deficient metabolic gene as genetic markers to restore wildtype characteristics. Three separate F. graminearum strains with two essential gene deletions were generated and validated.
